Effects of Hepatocyte on DNA Synthesis Induced by Various Growth Factors in Mouse Primary Hepatocyte Culture



Abstract
To test the effects of various cheimcal on mouse heaptocyte, we try to set up the mouse hepatocyte culture system from ICR mouse. The conditions of hepatocyte were investigated with biochemietiy and morphology. Based on studies with hepatocyte culture in 24hr conditions after culture, the hepatocyte growth factor substances EGF (Epidermal growth factor), TGF ¥á(Transformaling frowth factor alpha) have been defined as complete mitogens for mouse hepatocytes and implicated in control of liver growth. Cultured mouse hepatocytes were not treated with any growth factors without standard substances and William E medium, and time sequence difference were seen with morphology.
After finding the beat conditions in mouse hepatocyte culture, they were exporsed to EGF(2.5, 5, 10, 20, and 40ng/§¢) and TGF-a(2.5, b, 10, 20, and 40ng/§¢), and we determined DNA synthesis by measuring BrdU incorpoation into extracted DNA or by deterring the positive labeling index by means of immunohistochemistry. The resole show that EGF, TGF-¥á have strong mitogenic activity for mouse hepatocyte, although it showed difference of potent following dose. The level of mitogenic effects on mouse hepatocyte was higher TGF-¥á better than EGF ranging from 1.5- to 2.2 fold, depending on the experiment.
